InVitro+Jobs
stellt regelmäßig Wissenschaftler und ihre innovativen Forschungen als „Arbeitsgruppe im Portrait“ vor. Im Fokus stehen neu entwickelte Methoden, ihre Evaluation sowie der Ausblick, welche tierexperimentellen Versuchsansätze gemäß dem 3R-Prinzip (reduce, refine, replace) nach Möglichkeit reduziert und bestenfalls ersetzt werden können.  

In dieser Ausgabe thematisieren wir eine wichtige Forschungsarbeit von Prof. Dr. Gerhard Gstraunthaler, Leiter der Arbeitsgruppe „Molekulare Physiologie der Niere“ an der Medizinischen Universität Innsbruck. Er hat eine humanspezifische Methode entwickelt, mit der das fetale Kälberserum, ein Bestandteil in fast allen Nährmedien in der Zellkultur, ersetzt werden kann.

1. WORKING GROUP – A PORTRAIT: „Renal Molecular Physiology“ at the Medical University of Innsbruck

InVitro+Jobs presents scientists and their innovative research in a regular feature called “Working Group – a Portrait”. We focus on newly developed methods, their evaluation and their potential for reducing and where possible replacing animal experimentation according to the 3R principles of Russel & Burch (reduce, refine, replace).

In this edition we feature important research conducted by Prof. Dr. Gerhard Gstraunthaler, head of the working group „Renal Molecular Physiology“ at the Medical University of Innsbruck. The working group investigates questions regarding the biochemistry and physiology of the kidney and mostly works with epithelial cell and tissue cultures. A further focus is on replacement methods for animal experiments. We would like to emphasise a human-specific method with which the cell physiologist Prof. Gstraunthaler and his team intend to replace foetal bovine serum (FBS), a component of almost all nutrient media for cell cultures.

2. SPECIES DIFFERENCES: N-Acetylneuraminsäure versus N-Glycolylneuraminsäure

Sialinsäuren (Sia) kommen auf den Zelloberflächen bei allen Wirbeltieren und höheren Wirbellosen vor und sind essentiell u.a. bei der Embryonalentwicklung. Mit Hilfe dieser Oberflächenproteine sowie eines Komplementfaktors H und Sia-bindender Ig-ähnlicher Lektine kann das Immunsystem körpereigene von körperfremden Molekülen unterscheiden.

Beim Tier kommt die N-Glycolylneuraminsäure (Neu5Gc) vor, beim Menschen dagegen die N-Acetylneuraminsäure (Neu5Ac). Sie ist Hauptbestandteil von Glykolipiden, Glyko-proteinen und Proteoglykanen. Untersuchungen haben gezeigt, dass im Laufe der Evolution des Menschen im Vergleich zu unseren nächsten Verwandten, dem Menschen-affen, rund 10 verschiedene Gene verändert worden sind. Dieser humanspezifische Unterschied wird durch die Deletion eines wichtigen Exons im Umfang von 92 Basen-paaren im CMAH-Gen erklärt. Durch Verwendung von fötalem Kälberserum (FBS) besteht die Gefahr, dass z. B. humane therapeutische Stammzell-Linien die nur bei Tieren vorkommende Sialinsäure (N-Glycolyl-Neuraminsäure, Neu5Gc) in die Zellmembran einbauen. Die vom Tier stammende Sialinsäure kann u.a. die "Serum sickness reactions" verursachen, bei der das menschliche Immunsystem auf die aus tierischen Seren stammende Neu5Gc reagiert.

2. SPECIES DIFFERENCES: N-Acetylneuraminic acid vs. N-Glycolylneuraminic acid

Sialic acids (Sia) are present on the cell surfaces in all vertebrates and higher invertebrates. They are essential, amongst other things, for embryonic development. With the help of these surface proteins together with the complement factor H and Sia-binding Ig-like lectins, the immune system can distinguish endogenous from exogenous molecules.

Animals express the N-glycolylneuraminic (Neu5Gc), humans the N-acetylneuraminic acid (Neu5Ac). It is the main constituent of glycolipids, glyco-proteins and proteoglycans. Studies have shown that approximately 10 different genes have been altered in the course of human evolution in comparison with our closest relatives, the primates. This human-specific difference is explained by the deletion of an important exon with 92 bases pairs in the CMAH gene. Using foetal bovine serum (FBS) imvolves the risk of human therapeutic stem cell lines incorporating the sialic acid (N-glycolyl neuraminic, Neu5Gc) from the animal serum in their own cell membrane. The animal sialic acid may, amongst other things, cause “serum sickness reactions”, in which the human immune system responds to the Neu5Gc from animal sera.

München: iPS-Zentrum CARE geht 2017 an den Start

In München wird in den kommenden Jahren das Stammzelltechnologie-Zentrum „Center for Regenerative Engineering“ CARE errichtet. 15 Millionen Euro Anschubfinanzierung hat die bayerische Wirtschaftsministerin Ilse Aigner Medienberichten zufolge dem Initiator, dem Stammzellforscher Prof. Hans Schöler aus Münster für den Aufbau des Translations-zentrums zugesagt.
CARE kombiniert moderne Stammzelltechnologien mit der Pharmaforschung. Mithilfe von iPS-Zellen und Krankheitsmodellen in der Petrischale sollen hier Wirkstoffe im großen Maßstab getestet werden. So könnte eine Vielzahl an Tierversuchen wegfallen.

Organ-on-a-chip: Neuer Mechanismus von Paracetamol entdeckt

Ein israelisch-deutsches Forscherteam unter der Leitung von Prof. Yaakov Nahmias vom Alexander Grass-Center für Bioengineering an der Hebrew University in Jerusalem hat ein Organ-on-a-Chip-Modell mit einer winzigen Humanleber entwickelt, die mehr als einen Monat lebensfähig ist. Da sie in die Zellen zusätzlich optoelektrische Sensoren einbauten, gelang es ihnen, eine wichtige Entdeckung zur Toxizität von Acetaminophen zu machen – in Deutschland bekannt als Paracetamol.

DFG fördert Lab-on-a-Chip-Forschergruppe
Die Leipziger Forschergruppe „Integrierte chemische Mikrolaboratorien“ wird in den nächsten Jahren mit einer Forschungsförderung der Deutschen Forschungsgemeinschaft unterstützt.
Ziel sei es, chemische Synthesen aus den Chipsystemen im Mikromaßstab mit den Möglichkeiten miniaturisierter Analytik zu verknüpfen und diese Möglichkeiten auszubauen, hieß es in einer aktuellen Pressemitteilung der DFG dazu.
Mit Hilfe der Mikrolaboratorien könnten zum Beispiel neue pharmazeutische Wirkstoffe entwickelt und der Energie- und Ressourcenverbrauch chemischer Prozesse im Hochdurchsatz minimiert werden.

Hessen: Drei Professuren für weniger Tierversuche
Mit zwei neuen Professuren sowie der Wiederbesetzung einer Professur will die hessische Landesregierung zum Abbau der Tierversuche beitragen. Sie unterstützt dieses Projekt an zwei Standorten - Frankfurt und Gießen - mit 2 Millionen Euro in den nächsten fünf Jahren.
Die neue Professur an der Goethe-Universität Frankfurt ist für die Entwicklung tierversuchs-freier Methoden vorgesehen, um neue Wirkstoffe zu prüfen.
An der Veterinärmedizinischen Fakultät der Justus–Liebig Universität Gießen wird eine nicht besetzte Tierschutz-Professur neu besetzt und zusätzlich eine Juniorprofessur zur Förderung von Nachwuchswissenschaftlern eingerichtet. Die Gießener Professuren werden sich sowohl mit der Leidensreduktion in Tierversuchen als auch mit tierversuchsfreien Methoden beschäftigen.

Munich: iPS centre CARE starts in 2017
In the following two years, the stem cell technology center “Center for Regenerative Engineering” (CARE) will be built in Munich. According to media reports, the Bavarian Bavarian State Minister of Economic Affairs, Ilse Aigner, has pledged 15 million euros start-up funding for the stem cell researcher Prof. Hans Schöler to set up the translational centre. CARE combines modern stem cell technologies with pharmaceutical research. Drugs can be tested on a large scale with the help of iPS cells and disease models in the petri dish, which could render many animal experiments unnecessary.

Organ-on-a-chip: New mechanism of paracetamol discovered
An Israeli-German research team headed by Prof. Yaakov Nahmias from the Alexander Grass Center of Bioengineering at the Hebrew University in Jerusalem has developed an organ-on-a-chip model with a minute human liver that is usable longer than a month. Since they also inserted opto-electronic sensors into the cells, they were able to make an important discovery regarding the toxicity of acetaminophen, also known as paracetamol.
The long-term toxicity of acetaminophen is mediated by the xenobiotic enzymes CYP2E1 and CYP3A4. The toxic metabolite N-acetyl-p-benzoquinonimin (NAPQI) causes protein adducts, which lead, among other things, to a damaging effect on the mitochondria impairing the cell respiration. In addition, an inflammatory response is triggered.

DFG promotes lab-on-a-chip research group
In the next few years, the Leipzig research group “Integrated chemical micro laboratories” will receive research funding from the German Research Foundation (DFG).
The goal is to combine chemical syntheses from micro-scale chip systems with the possibilities of miniaturised analytics and expand these possibilities, according to a recent press release by the DFG.
For instance, with the help of the micro laboratories, new pharmaceutical agents could be developed and the energy and resource consumption of chemical processes in high throughput procedures could be minimised.

Hesse: Three Professorships for less animal testing
The Hessian state government intends to contribute to the reduction of animal experiments with two new professorships and the reoccupation of a further professorship. It will fund this project at two sites, Frankfurt and Giessen, with 2 million euros during the next five years.
A new professorship for the development of animal-free methods for testing new drugs is planned at Goethe University in Frankfurt.
In the Faculty for Veterinary Medicine at Justus-Liebig University of Giessen a vacant animal welfare professorship will be occupied. Additionally, a junior professorship will be established to promote young scientists. Both Giessen professors will address the reduction of animal suffering in animal experiments as well as animal-free methods.
